How we study it

Techniques and Analyses

We use a combination of viral methods and molecular tools to perturb, manipulate, and visualize brain activity along with cell-specific imaging:

  • in-vivo single-cell calcium imaging (miniscopes)
  • chemogenetics
  • optogenetics
  • immunohistochemistry

We model behavioral inflexibility and drug taking using gold-standard preclinical models:

  • Intravenous self-administration of methamphetamine
  • 2-bottle choice alcohol drinking

We partner these techniques with custom-designed behavioral paradigms combined with a variety of analytical methods:

  • Generalized Linear Models (GLMs)
  • Machine Learning (SVM decoding)
  • Motion and postural tracking

to decode and better understand brain and behavior relationships

Heatmap of over 300 orbitofrontal cortex neurons, one row per cell sorted by peak firing time, activity aligned to choice time: a bright band of sequential peaks sweeping from before the choice to about three seconds after it.
Peri-event time histogram of orbitofrontal cortex neurons responsive to reward, collected by Dr. JL Romero Sosa.
Occupancy figure from overhead video tracking of a rat in a behavioural arena: thousands of yellow keypoint detections and teal movement traces accumulated across a session, plotted over a frame of the arena.
